No you can play whenever. If you get your 50 hours before the game calculates your played in-game time then you should get your ribbon at that point. If you don't get your 50 hours and are still playing between 4am and 6am on any given day you may have to wait until the next day. -

Another ribbon "easy" to get, nice of you.
Just to be sure, 50 hours of game needs all the usual conditions (at least 6 players at the end of the game, game finished i.e. no quitting or kicking counting, etec.) ?
should be in game all day long
